Alpha-Lipoic Acid Downregulates IL-1β and IL-6 by DNA Hypermethylation in SK-N-BE Neuroblastoma Cells

چکیده
Alpha-lipoic acid (ALA) is a pleiotropic molecule with antioxidant and anti-inflammatory properties, of which the effects are exerted through the modulation of NF-kB. This nuclear factor, in fact, modulates different inflammatory cytokines, including IL-1b and IL-6, in different tissues and cell types. We recently showed that IL-1b and IL-6 DNA methylation is modulated in the brain of Alzheimer's disease patients, and that IL-1b expression is associated to DNA methylation in the brain of patients with tuberous sclerosis complex. These results prompted us to ask whether ALA-induced repression of IL-1b and IL-6 was dependent on DNA methylation. Therefore, we profiled DNA methylation in the 5'-flanking region of the two aforementioned genes in SK-N-BE human neuroblastoma cells cultured in presence of ALA 0.5 mM. Our experimental data pointed out that the two promoters are hypermethylated in cells supplemented with ALA, both at CpG and non-CpG sites. Moreover, the observed hypermethylation is associated with decreased mRNA expression and decreased cytokine release. These results reinforce previous findings indicating that IL-1b and IL-6 undergo DNA methylation-dependent modulation in neural models and pave the road to study the epigenetic mechanisms triggered by ALA.

FIVE ALPHA DIHYDROTESTOSTERONE (5α-DHT) MAY MODULATE NITRIC OXIDE RELEASE VIA ENDOGENOU S CYTOKINES IN PERITONEAL MA CROPHA GES OF NZB/BALBc MICE
Recent studies have established that sex hormones directly or indirectly affect T and B cells and macrophages by manipulating the production of cytokines. In this study the possibility of the effect of 5a-DHT on macrophage (MΦ) nitric oxide (NO) release via interleukin-l, 6 (lL-1β, IL-6) or tumor necrosis factor-a (TNFα) was investigated. The endogenous cytokines IL-1β, IL-6 and TNF-α were ...

The pathophysiology of acute spinal cord injury (SCI) involves primary and secondary mechanisms of injury. Secondary injury mechanisms include inflammation, oxidative stress. The secondary inflammation of spinal cord tissue after SCI was critical for the survival of motor neuron and functional recovery. Flaxseed is a rich source of lignan phytoestrogen, α-linolenic acid. Flaxseed has rema...
متن کاملSalvianolic acid B improves insulin secretion from interleukin 1β-treated rat pancreatic islets: The role of PI3K-Akt signaling
Background and Objective: Oxidative stress induced by proinflammatory cytokines such as IL-1β plays a major role in β-cell destruction in diabetes type 1. Salvianolic acid B (Sal B) is a polyphenolic compound with antioxidant and protective effects. Thus, objective of this study was to assess the protection exerted by Sal B on isolated rat islets exposed to IL-1β and to investigate an underlyin...
متن کاملIncreased Levels of AIM2 and Circulating Mitochondrial DNA in Type 2 Diabetes
Background: Chronic inflammation has critical role in Type 2 diabetes (T2D), in which IL-1β contributes in insulin resistance and beta cell dysfunction. The activation of NLRP3 and AIM2 by endogens ligands, such as mtDNA can lead to the release of active form of IL-1β. Objective: To evaluate AIM2 expression and activation as well as circulating mtDNA levels in T2D patients. Methods: AIM2 expres...

Neuroblastoma resistance to apoptosis may contribute to the aggressive behavior of this tumor. Therefore, it would be relevant to activate endogenous cellular death mechanisms as a way to improve neuroblastoma therapy. We used the neuroblastoma SH-SY5Y cell line as a model to study the mechanisms involved in acetaminophen (AAP)-mediated toxicity by measuring CYP2E1 enzymatic activity, NFkB p65 ...
